Reconstitution is where most research error is introduced. The variables that matter are the diluent, the volume, the transfer technique and the storage that follows.

Choosing the diluent

Bacteriostatic water preserved with 0.9% benzyl alcohol is used for multi-draw vials. Sterile water without a preservative is single-use and should be discarded after the first draw.

The concentration maths

Concentration is milligrams of peptide divided by millilitres of diluent. A 10 mg vial reconstituted with 3 ml gives 3.33 mg/ml; the same vial with 2 ml gives 5 mg/ml.

Transfer technique

Direct the stream of water down the inner wall of the vial rather than onto the powder cake. Swirl gently until clear — never shake, as agitation shears peptide chains.

After reconstitution

Store refrigerated at 2–8 °C, protected from light, and log the reconstitution date on the vial. Undated vials are unusable data.
